The blue hedgehog and the red echidna were avowed opponents in Sonic 3 but in the platform exploit Sonic & Knuckles™, Knuckles comprehends that he's been just a wager in the schemes of Dr. Eggman (aka Dr. Robotnik). Knuckles the previous opponent now support Sonic to pursue the ordinary enemy and each must employ their strengths to obtain the invaluable Master Emerald and save Angel Island.
